根据已报道的序列，把苦瓜(Momordica charantia) MAP30基因成功克隆到原核表达载体pET28a（+）中，并对PCR快速检测阳性克隆进行了研究。结果表明，直接用菌落、菌液、酚氯仿处理过的菌液,以及提取的质粒进行PCR都可以成功地筛选阳性菌落。其中，酚氯仿处理过的菌液PCR与质粒PCR的结果最接近，而且比质粒PCR简单，因此可作为方便可靠的阳性克隆筛选的新方法。Abstr...

摘要本实验构建了含有CaMV35S启动子控制下的Pti5-VP16基因的植物双元表达载体pBI121UCH1。通过根癌农杆菌叶盘转化法，将Pti5-VP16基因导入烟草SRI中，经卡那霉素筛选，获得了抗性植株。经PCR和Southern印迹分析，表明抗性植株中整合了Pti5-VP16基因，经抗病性鉴定转基因烟草植株的抗病性明显提高。
